Purpose It is used to distinguish between thin collagen fibers and thick bundles.
Ingredients A) Picro Sirius Red Solution: Sirius red, Picric acid
B) Buffer Solution: Acetic acid
C) Hematoxylin Mayer's Solution: Ethanol 96%, Hematoxylin, Sodium iodate, Glycerol, Triton X-100
Working Principle Pikro Sirius Red Staining Kit, is used to identify and color connective tissue components. It is especially preferred for the demonstration of collagen fibers. The solution is applied after cutting and preparation of tissue samples. Staining collagen fibers red increases the visibility of these fibers under the microscope and facilitates the examination of tissue samples. The buffer solution preserves the structure of the tissue sample and prevents the loss of cellular details. Hematoxylin Mayer solution is used to stain the nuclei of tissue samples. Hematoxylin interacts with the nucleic acids (DNA and RNA) in the tissue sample and stains the nuclei blue. In this way, the cell nuclei in the tissue samples become clearer and can be viewed more clearly under a microscope.
Warnings, Precautions, Recommendations Danger. Causes severe skin burns and eye damage. Wear protective gloves/protective clothing/eye protection/face protection. If swallowed: Rinse mouth. DO NOT attempt to void. IF IN CONTACT WITH SKIN (or hair): Immediately remove all contaminated clothing. Rinse skin with water/shower. IN CASE OF CONTACT WITH EYES: Rinse carefully with water for several minutes. Remove contact lenses if fitted and easy to do so. Continue rinsing. Immediately call NATIONAL POISON ADVICE CENTER PHONE 114 or doctor/physician. Keep under lock and key.
Application 1.Add 10 drops of “A-Picro Sirius Red Solution” to the preparation and wait 50 minutes.
2.Rinse 2 times with “B- Buffer Solution”.
3.Add 10 drops of “C- Hematoxylin Mayer Solution” and wait for 5 minutes.
4.Wash under running tap water for 5 minutes.
5.Dehydrate in ethyl alcohol.
6.Clean in xylene and cover.
