Purpose
Used to demonstrate acid epithelial mucin (sialomucin, sulfomucin) and stromal (mesenchymal) mucin.
Ingredients
A) Acetic Acid Aqueous Solution: Glacial Acetic Acid
B) Colloidal Iron Stock Solution: Iron (III) Chloride
C) Acetic Acid, Glacial: Glacial Acetic Acid
D) Potassium Ferrocyanide Solution: Potassium Ferrocyanide Trihydrate
E) Hydrochloric Acid Solution: 37% Hydrochloric Acid
F) Van Gieson Solution: Picric Acid, Acid Fuchsin
Working Principle
Colloidal Iron Acid; Mucin Staining is a technique used to histologically identify mucus and acidic mucopolysaccharides (mucin). This method is an important tool for identifying and examining mucus-containing cells and tissues. Colloidal iron staining is based on the binding of acidic mucopolysaccharides and mucus to iron ions. Once bound to acidic mucopolysaccharides, iron is visualized by a Prussian blue reaction.
Warnings, Precautions, Recommendations
Danger. Flammable liquid and vapour. May cause cancer. Causes severe skin burns and eye damage. Wear protective gloves/protective clothing/eye protection/face protection. IF exposed or concerned: get medical advice/attention. IF SWALLOWED: rinse mouth. DO NOT induce vomiting. IF ON SKIN (or hair): remove/take off immediately all contaminated clothing. Rinse skin with water/shower. IF IN EYES: Rinse cautiously with water for several minutes. Remove contact lenses, if present and easy to do. Continue rinsing. Causes skin irritation.
Application
Pre-Staining Preparation:
1. It is recommended to use clean glassware in the staining procedure.
2. Preparation of Colloidal Iron Working Solution: Add and mix.
a. B-Colloidal Iron Stock Solution 20 ml
b. C-Acetic Acid, Glacial 5 ml
c. Distilled Water 15 ml
Staining:
3. Deparaffinize the sections by placing them in xylene for 3 minutes. Repeat this process 3 times. Then dip them in 100% and 96% denatured ethyl alcohol twice. Wash thoroughly with distilled water.
4. Place in Solution A (Acetic Acid Solution, 12%) for 30 seconds.
5. Dry the slides. Do not wash.
6. Place in Colloidal Iron Working Solution for 30 minutes.
7. Wash 3 times with Solution A (Acetic Acid Solution, 12%) for 3 minutes each.
8. Prepare Ferrocyanide-Hydrochloric Acid Solution just before use. Add and mix well.
a. Solution D: Potassium Ferrocyanide Solution, 2% 20 ml
b. Solution E: Hydrochloric Acid Solution, 2% 20 ml
9. Place in Ferrocyanide-Hydrochloric Acid Solution for 15 minutes.
10. Wash in tap water for 5 minutes.
11. Counterstain in Solution F: Van Gieson Stain for 3-5 minutes.
Continue directly to dehydration step without rinsing.
12. Dip twice in 100% denatured ethyl alcohol, dry. Then dip the slide in xylene three times and cover the slide.
