Purpose
In histology, histopathology and clinical pathology, the Perls Prussian Blue VanGieson Staining Set is used to detect the presence of ferric iron containing hemosiderin in tissue or cell samples.
Ingredients
A) Potassium Ferrocyanide Solution: Potassium Ferrocyanide.
B) Hydrochloric Acid Solution: 37% Hydrochloric acid.
C) VanGieson's Picro-Acid-Fucsin Solution: Picric acid, Fucsin
Working Principle
Ferric iron deposits in the tissue (mostly present as ferric iron in the storage protein ferritin) react with the soluble ferrocyanide in the dye to form the insoluble Prussian Blue pigment. These deposits are then visualized microscopically as blue or purple deposits.
Warnings, Precautions, Recommendations
Danger Causes severe skin burns and eye damage. Wear protective gloves/protective clothing/eye protection/face protection. If swallowed: Rinse mouth. DO NOT attempt to void. IF IN CONTACT WITH SKIN (or hair): Immediately remove all contaminated clothing. Rinse skin with water/shower. IN CASE OF CONTACT WITH EYES: Rinse carefully with water for several minutes. Remove contact lenses if fitted and easy to do so. Continue rinsing. INHALATION: Remove the affected person to fresh air and keep in a comfortable position so that he/she can breathe easily.
Application
Incubation Solution
Take 0.5 ml of “A - Potassium Ferrocyanide Solution”, 0.5 ml of “B - Hydrochloric Acid Solution” and mix with 1.5 ml of distilled water. Leave for 10 minutes. Mix again. Use fresh.
1. Cover with 10 drops of “Incubation Solution”, incubate for 30 minutes, then shake the slide to remove the reagent.
2. Wash with distilled water for 2 minutes.
3. Cover with 10 drops of “C- VanGieson's Picro-acid-fucsin Solution”. After 10 minutes remove the reagent by shaking the slide.
4. Dehydrate, clean, cover with synthetic resin and examine under a microscope.
