Purpose |
It is the most commonly used staining method in gynecological and non-gynecological cytological smears. PAP (Papanicolaou stain) is the main staining method for SVS and is widely used in all other types of cytologic material. With the regressive method, nuclei are overstained and this excess is removed with an acid-alcohol solution. |
Ingredients |
A) Hematoxylin Harris Solution: Hematoxylin, Ethylene glycol, Glycerol, dimethyl sulfoxide, Acetic acid
B) Acid Alcohol Solution: Hydrochloric acid, Ethanol
C) Papanicolaou, OG-6 Stain: Methanol, Propanol, Orange G dye
D) Papanicolaou, EA-50 Stain: Propanol, Methanol, Eosin Y, Light green SF stain |
Working Principle |
The use of stains such as hematoxylin and eosin allows the different structures of cells, such as both their nuclei and cytoplasm, to be highlighted. This allows for a more comprehensive assessment of cellular features and allows for an accurate diagnosis of the disease. |
Warnings, Precautions, Recommendations |
Caution Danger Highly flammable liquid and vapor. Causes skin irritation. Causes damage to organs. Causes eye irritation. Keep away from heat/sparks/flame/hot surfaces. - No smoking. Wear protective gloves/protective clothing/eye protection/face protection. Harmful if swallowed. Causes organ damage after prolonged or repeated exposure (kidney). Do not breathe dust/fume/gas/mist/vapor/spray. If you feel unwell, seek medical advice/attention. IF SWALLOWED: If you feel unwell, call NATIONAL POISON ADVICE CENTER PHONE 114 or a doctor/physician. IF IN CONTACT WITH SKIN (or hair): Immediately remove all contaminated clothing. Rinse skin with water/shower. If eye irritation persists: Get medical advice/attention. Store in a well ventilated area. Keep cold. |
Application
|
1. Soak the smear in 95% ethanol for 15 minutes.
2. Immerse 10 times in tap water (hydration).
3. Stain in A-Hematoxylin Harris Solution for 2 minutes.
4. Immerse 10 times in tap water.
5. B- Immerse quickly several times in Acid Alcohol Solution and remove. (The nucleus should become clear and the ground should be bright or colorless.)
6. Immerse 10 times in tap water.
7. Immerse 10 times in 95% ethanol (dehydration). Repeat 2-3 times in separate bottles.
8. Stain in C- Papanicolaou, OG-6 Stain for 2 minutes.
9. Immerse 10 times in 95% ethanol. Repeat 2-3 times in separate bottles.
10. Stain in D- Papanicolaou, EA-50 Stain for 5 minutes.
11. Immerse 10 times in 95% ethanol. Repeat 2-3 times in separate bottles.
12. Dry.
13. Dip 20 times in Xylol.
14. Cover with entellan before xylene dries. |
