Purpose Used for rapid detection of eosinophils in smears and tissue sections.
Ingredients A) Eosin Y Alcoholic Solution 0.5%: Eosin Y, Methanol, Dimethyl sulfoxide, Glycerol
B) Methylene Blue Solution 1%: Methylene blue, Methanol, Dimethyl sulfoxide, Glycerol
C) Phosphate Buffer Solution pH 6.8: Phosphate Buffers, Preservative
D) Methanol Methanol.
Working Principle Hansel's Eosinophil Staining Kit is a mixture of methylene blue and eosin Y. It is a specialized staining method used to identify eosinophil cells in urine and other body fluids.
Warnings, Precautions, Recommendations Toxic Easily flammable. Danger of very serious, incurable effects by ingestion, skin contact, inhalation. Causes irritation to eyes. May have an irritant effect on the skin at high concentrations and prolonged exposure. In case of inhalation: Vapors may cause drowsiness and dizziness. If swallowed: May cause damage to the mouth and larynx. A doctor should be consulted. Ingestion of chemicals is a health hazard.
Application SAMPLE AND PREPARATION:
Eosin Buffered Solution
Take a suitable container. Add 25 ml of “A- Eosin Y Alcoholic Solution 0.5%” and 25 ml of “C- Phosphate Buffer Solution 6.8”. Close the lid. Stir. Wait 10 minutes. Mix again.

Methylene blue buffered solution
Take a suitable container. Add 25 ml of “B-Methylene Blue Solution 1%” and 25 ml of “C-Phosphate Buffer Solution 6.8”. Close the lid. Stir. Wait 10 minutes. Mix again.

STAINING METHOD:
1. Immerse in a container of A-Eosin Y Alcoholic Solution 0.5%. After 1 minute, shake off.
2. Immerse in a container of Eosin buffered solution. After 1 minute, shake off.
3. Wash briefly with running distilled water.
4. Immerse in a beaker of D- Methanol. Shake off after 15-20 seconds.
5. B- Immerse in a beaker of Methylene Blue Solution 1%. Shake off after 1 minute.
6. Immerse in a beaker of methylene blue buffered solution. Shake off after 1 minute.
7. Immerse in a beaker of D- Methanol. Shake off after 15-20 seconds.
8. Air dry.
9. Examine under a microscope.
