Purpose
Congo Red Highman is a histological staining method used specifically to detect amyloid deposits.
Ingredients
A) Congo Red Solution: Ethanol 96%, Congo red
B) Alkaline Solution: Ethanol 96%, Potassium buffers
C) Phosphate Buffer Solution: Phosphate buffers
D) Mayer Hemalum Solution: Hematoxylin, Aluminum ammonium sulfate, Glycerol
Working Principle
Congo Red: Congo red is an aniline dye that has a tendency to bind to amyloid proteins. This binding allows amyloids to be clearly visible under the microscope. Sections are incubated in Congo red solution for a specific time. In this step, Congo red binds to amyloid fibrils, causing the amyloid deposits to appear red.
Warnings, Precautions, Recommendations
Danger. Warning. Highly flammable liquid and vapour. Causes serious eye irritation. May cause cancer. Keep away from heat/sparks/flames/hot surfaces. - No smoking. Keep container tightly closed. IF ON SKIN (or hair): Remove/Take off immediately all contaminated clothing. Dry skin with water/shower. IF IN EYES: Rinse cautiously with water for several minutes. Remove contact lenses, if present and easy to do. Continue rinsing.
Application
1. Cover the preparation with 10 drops of “A- Congo Red Solution”.
2. After 15 minutes, shake the slide to remove the reagent.
3. Wash with distilled water for 1 minute.
4. Cover the preparation with 10 drops of “B- Alkaline Solution”.
5. After 30 seconds, shake the reagent
6. Wash with distilled water for 5 minutes.
7. Cover the preparation with 10 drops of “C- Phosphate Buffer Solution”.
8. After 2 minutes, shake the slide to remove the reagent.
9. Cover with 10 drops of “D- Mayer Hemalum Solution” without washing.
10. After 5 minutes, shake the slide to remove the reagent.
11. Wash with distilled water for 5 minutes.
12. Dehydrate, clean, cover with synthetic resin and examine under a microscope.
