Purpose
It is a hematoxylin solution used in histology and cytology, particularly for nuclear staining.
Ingredients
Hematoxylin-approved (CI-75290), Sodium iodate, Aluminum ammonium sulfate, Acetic acid, Preservatives and Stabilizers.
Working Principle
Hematoxylin (CI-75290); when oxidized, it forms hematin, which is blue-violet in color. Thanks to this property, it is used as a suitable stain fixative for staining cell nuclei. It facilitates the examination of transparent cells and tissues under a microscope.
Warnings, Precautions, Recommendations
Danger. Causes serious eye damage. Prolonged or repeated exposure may cause organ damage. Wear protective gloves/protective clothing/eye protection/face protection. IF IN EYES: Rinse cautiously with water for several minutes. Remove contact lenses, if present and easy to do. Continue rinsing. Immediately call the NATIONAL POISON CONTROL CENTER at 114 or your doctor/physician. If you feel unwell, seek medical advice/attention. If eye irritation persists: Seek medical advice/attention.
Application
1. Tissue sections are prepared.
2. Deparaffinization and rehydration are performed.
3. The specimen is immersed in “Hematoxylin Stain, Gill 2”.
4. The staining time is typically 1–5 minutes (depending on the protocol).
A longer time may result in darker nuclear staining.
5. Slides are washed in tap water for 2–5 minutes.
This washing also promotes color development (bluing). Alternatively, bluing can be performed using “GBL® Ref 2210- Bluing Solution.” This is followed by a brief wash with distilled water.
6. Counterstaining can be performed. For example, using “GBL® Ref 5014-Hematocystin & Eosin Staining Kit.”
7. Dehydration (slides are dehydrated using a series of ethanol solutions: 70% → 96% → 100%).
8. Clarification (xylene, etc.)
9. The prepared slide is left to dry completely at room temperature (30 °C in an oven).
10. It is examined under a microscope using a 100x objective.
