Purpose |
It is the most commonly used staining method for gynecologic and non-gynecologic cytologic smears. PAP (Papanicolaou stain) is the main staining method for SVS and is widely used in all other types of cytologic material. The method used is the progressive method. In the progressive method, nuclei are stained at the desired intensity. |

A) Hematoxylin Solution; Gill's: Hematoxylin, Ethylene Glycol, Acetic acid, Aluminum sulfate, Sodium iodate
B) Blueing Solution: Ammonium hydroxide

D) Papanicolaou, EA-50 Stain: Methanol, Bismark brown, Acetic acid, Eosin Y, Light green SF stain, Ethanol, 96 |
Working Principle |
The Pap staining kit includes a specific staining protocol and is designed to highlight specific components of cervical smear samples. The GILL'S staining set can be used in a variety of staining protocols and contains different staining solutions. These solutions are designed to characterize different components of cells and tissues. |
Warnings, Precautions, Recommendations |
Caution Danger Highly flammable liquid and vapor. Toxic if swallowed, in contact with skin or inhaled.
Causes damage to organs. Causes skin irritation. Causes serious eye irritation. Keep away from heat/sparks/flame/hot surfaces - Do not smoke. Prolonged or repeated exposure may cause damage to organs. Do not breathe dust/fume/gas/mist/vapor/spray. Wear protective gloves/protective clothing/eye protection/face protection. IF IN CONTACT WITH SKIN (or hair): Immediately remove all contaminated clothing. Rinse skin with water/shower. IN CASE OF CONTACT WITH EYES: Rinse carefully with water for several minutes. Remove contact lenses if fitted and easy to do so. Continue rinsing. In case of exposure: Call NATIONAL POISON ADVISORY CENTER TELEPHONE 114 or doctor/physician. |
Application
|
1. Fix in 95% ethanol for 15 minutes.
2. Immerse 10 times in tap water (Hydration)
3. Stain in A-Hematoxylin Solution Gill's for 1 minute.
4. Immerse 10 times in tap water.
5. Soak in B- Blueing Solution for approximately 30 seconds. (Sections are immersed until bright blue)
6. Immerse 10 times in tap water.
7. Immerse 10 times in 95% ethanol (dehydration). Repeat 2-3 times in separate bottles.
8. Stain in C- Papanicolaou, OG-6 stain for 2 minutes.
9. Immerse 10 times in 95% ethanol. Repeat 2-3 times in separate bottles.
10. Stain in D- Papanicolaou, EA-50 Stain for 5 minutes.
11. Immerse 10 times in 95% ethanol. Repeat 2-3 times in separate bottles.
12. Dry.
13. Dip 20 times in Xylol.
14. Cover with entellan before xylene dries. |
