Purpose
It is used for counting spermatozoids.

Working Principle
Formaldehyde is a fixative commonly used in spermatozoid counting solution. Fixatives preserve the structure and properties of cells, preventing them from changing during laboratory analysis. Formaldehyde is particularly known for its ability to fix cells, which helps to maintain the morphology (shape and structure) of sperm cells throughout laboratory studies.Sodium hydrogen carbonate (NaHCO₃) is a compound used in spermatozoid counting solution and is added to regulate the pH level of the solution. This helps to prevent the solution from becoming acidic or basic and allows spermatozoa to perform better under laboratory conditions.
Warnings, Precautions, Recommendations
Not classified as hazardous according to the Regulation on SEA of Substances or Mixtures (OG 11.12.2013 No: 28848 bis) and Directive 1272&2008 [CLP/GHS](EC).
Application
1.Use small volume containers and work aseptically to avoid contamination of the solution.
2.Seminal fluid is used directly for sperm counting.
3.Take a leukocyte counting pipette with a white bead inside.
Withdraw seminal fluid up to the -0.5 mark.
4.Add working solution to the 11 mark.
5.Mix properly.
6.Care should be taken to avoid the formation of air bubbles.
-3-4 drops of the mixture are poured out.
7.Then add 1-2 drops to the Neubauer counting chamber.
8.Cover with a coverslip and examine under a microscope.
Theoretical Results
Using a microscope at 300x magnification, 1 mm2 areas in each corner are counted (i.e. 4 areas are counted).
The result is multiplied by 50. This is the number of spermatozoids per 1 mm3.
When this number is multiplied by 1,000, the number in 1cm3 is found.
