Purpose
It is usually used for staining blood cells.
Ingredients
Giemsa stain, Methanol and Glycerol.
Working Principle
Giemsa stain is a differential stain and contains a mixture of azure, methylene blue and eosin stain. It is specific for the phosphate groups of DNA and binds itself where there is a high amount of adenine-thymine bonding. Azur and eosin are acidic dyes that variably stain the basic components of cells such as cytoplasm, granules, etc. Methylene blue, on the other hand, acts as a basic stain for acidic components, especially the cell nucleus.
Warnings, Precautions, Recommendations
Highly flammable liquid and vapour. Toxic if swallowed, in contact with skin or inhaled. Causes damage to organs.
Application
Staining of Smears
1. Preparation of Dye Solution (Dilute Giemsa Solution):
▪ Take 5 ml of GBL-2408 "Giemsa Stain; Concentrate" into the staining container.
Add 45 ml of distilled water and mix well.
2. Staining with Giemsa Solution:
▪ Cover the preparation with methanol and fix for 5 minutes.
Immerse the preparation in the prepared staining solution.
After 15-30 minutes, remove the slide and shake to remove the reagent.
Wash with distilled water. Dry it.
Examine under a microscope.
