Purpose
It is used to demonstrate calcium stores.
Ingredients
A)Silver Nitrate Solution 5%: Silver nitrate
B) Sodium Thiosulfate Solution 5%: Sodium thiosulfate
C) Nuclear red solution 0.1%: Aluminum sulfate, Nuclear fast red
Working Principle
The Von Kossa Calcium Staining Kit is widely used in histology to detect the presence of abnormal calcium deposits in the body. The principle of this coloring is based on the conversion of calcium salts into silver salts. Calcium ions bound to phosphates are replaced by silver ions brought in with silver nitrate solution.
Warnings, Precautions, Recommendations
Danger Causes severe skin burns and eye damage. Very toxic in aquatic environment. Long lasting, very toxic effect in aquatic environment. Avoid release into the environment. Wear protective gloves/protective clothing/eye protection/face protection. IN CASE OF SKIN CONTACT: Wash with plenty of soap and water. IN CASE OF EYE CONTACT: Rinse carefully with water for several minutes. Remove contact lenses if fitted and easy to do so. Continue rinsing.
Application
1. Immerse in a container of A-Silver Nitrate Solution. After 60 minutes, shake off.
2. Wash with distilled water.
3. Immerse in a container of B-Sodium Thiosulfate Solution. Shake off after 3 minutes.
4. Wash with distilled water.
5. Immerse in a beaker of C-Core Red Solution. After 5 minutes, shake off.
6. Wash thoroughly with distilled water.
7. Dehydrate by passing through various alcohols or denatured alcohol substitutes.
8. Clean by passing through xylene or substitutes.
9. Cover with synthetic resin.
10. Examine under a microscope.
