Purpose
Used for manual counting of white blood cells.
Ingredients
Glacial acetic acid
Working Principle
The acetic acid in the solution breaks down the erythrocytes, allowing the leukocytes to be seen more clearly.
Warnings, Precautions, Recommendations
Danger. Causes severe skin burns and eye irritation. Wear protective gloves/protective clothing/eye protection/face protection. IF SWALLOWED: rinse mouth. DO NOT induce vomiting. IF ON SKIN (or hair): Remove/Take off immediately all contaminated clothing. Rinse skin with water/shower. IF INHALED: If breathing is difficult, remove victim to fresh air and keep at rest in a position comfortable for breathing. IF IN EYES: Rinse cautiously with water for several minutes. Remove contact lenses, if present and easy to do. Continue rinsing. Dispose of contents/container in accordance with national regulations.
Application
1. Take a white bead leukocyte counting pipette.
2. Blood is drawn up to the 1 mark. In cases where severe white blood cell increase is suspected, blood is drawn up to the 0.5 mark.
3. “Leukocyte Counting Solution, Colorless” (2694) is taken. This reagent is drawn up to the 11 mark. The pipette is shaken. A few drops are thrown out.
4. Take a Thoma counting chamber and drop 1-2 drops of the reagent-blood mixture in the pipette.
5. Cover the coverslip over the chamber.
6. Count all the leukocytes in the large square area with low magnification. The leukocytes on the left and upper lines are also included in the count. However, the leukocytes on the right and lower lines are not included in the count.
The total leukocyte count found is multiplied by 100. The result gives the total leukocyte count in 1 mm3.
Theoretical Results
The result gives the total number of leukocytes in 1 mm3.
