Purpose Gram staining method is used to stain gram negative bacteria.
Ingredients Safranine, methanol, triton X-100, ethanol, sodium azide.
Working Principle Gram staining bacteria are divided into Gram positive and Gram negative. This distinction is based on structural differences in the bacterial membrane of the two groups, i.e. their ability to retain the dye. GBL® CONVASTAIN Gram Safranine Stain is used for contrast staining of bacterial species that do not retain their primary stain, i.e. Gram negative bacteria.
Warnings, Precautions, Recommendations Flammable liquid and vapour. Harmful if swallowed. May cause damage to organs.
Application 1)If the smear is too acidic, cover with "Sodium Hydrogen Carbonate Solution" for 15-30 seconds.
2)Cover the smear with "GBL-2434 Crystal Violet Dye" for 1-2 minutes.
3)Wash with distilled water.
4)Cover the smear with "GBL-2435 Lugol's Solution" for 1 minute.
5)Washed with distilled water.
6)Cover smear with "GBL-2891 Denatured Alcohol" for 5-15 seconds.
7)Washed with distilled water.
8)Cover the smear with "GBL-2444 Gram Safranine Stain" for 1 minute.
9)Gently washed with distilled water, air dried and examined under a microscope.
