Purpose It is used in the examination of ferric ions and ferritin by light microscopy.
Ingredients A) Potassium Ferrocyanide Solution: Potassium Ferrocyanide
B) Acid Activation Solution: Hydrochloric acid
C) Nuclear Dark Red Solution: Aluminum sulfate, Nuclear fast red, Thymo
Working Principle Hemosiderin is an iron storage complex formed by partially digested ferritin and lysosomes. The breakdown of heme gives rise to biliverdin and iron. The body then captures the released iron and stores it in the tissues as hemosiderin. Cellular iron is found as ferritin or hemosiderin. In cells, it is identified by the Perls or Prussian blue reaction, in which ionic iron reacts with acid ferrocyanide to give a blue color. It appears blue under the microscope.
Warnings, Precautions, Recommendations Danger. May be corrosive to metals. Causes severe skin burns and eye damage. Keep only in original container. Wear protective gloves/protective clothing/eye protection/face protection. IF SWALLOWED: Rinse mouth. DO NOT induce vomiting. IF ON SKIN (or hair): Remove/Take off immediately all contaminated clothing. Rinse skin with water/shower.
Application Preparation of Incubation Solution:
Put the reagents of distilled water, A-Potassium Ferrocyanide Solution and B-Acid Activation Solution in a 50 mL container, respectively. Mix and wait for 10 minutes. Use fresh.
Application of Staining Method:
1. Immerse the preparation in the “Incubation Solution” prepared in the previous step. Incubate for 20 minutes at room temperature.
2. Wash with distilled water for 1 minute.
3. Cover the preparation with 10 drops of “C-Core Dark Red Solution”. After 5 minutes, shake the slide to remove the reagent.
4. Wash with distilled water for 1 minute.
5. Dehydrate, clean, cover with covering medium (GBL-2515) and examine under a microscope.
