Purpose It is used to detect mucins (mucopolysaccharides) in connective and supporting tissue cells (especially epithelium).
Ingredients A) Modified Mayer Hemalum Solution: Ethanol %96, Hematoxylin, Ammonium aluminum sulfate, Sodium iodate, Glycerol, Triton X-100, Citric acid
B)Mayer Mucicarmin Solution: Carmine, Aluminum hydroxide, Aluminum chloride, Ethanol %96
C)Methanil Yellow Solution: Methanil yellow, Glacial acetic acid
Working Principle Mucin is a secretion produced by various epithelial cells and connective tissue cells. In some intestinal inflammations or carcinomas, excessive amounts of mucin are secreted by epithelial cells. The mucicarmin staining technique is also useful for locating the primary tumor by visualizing mucin-producing tumor cells in an area that does not contain mucin-producing cells. Mucicarmin staining can also be used for staining encapsulated fungi such as cryptococci. The principle of mucicarmin staining is based on the presence of aluminum, which forms a chelating complex with carmine. This changes the charge of the carmine molecule to a positive charge, which allows it to bind to low-density acidic substrates such as mucins.
Warnings, Precautions, Recommendations Caution Dangerous. Harmful. May cause eye irritation. Prolonged inhalation may cause shortness of breath, mucosal irritation, coughing. Harmful if swallowed. May cause skin irritation after prolonged or repeated exposure.
Application 1. Cover the tissue section with 10 drops of “A-Modified Mayer Hemalum Solution” and shake the slide after 5 minutes to remove the reagent.
2. Wash with distilled water.
3. Add 0.5 ml of distilled water to the tissue section. Add 10 drops of “B-Mayer Mucicarmine Solution”. Mix by blowing and small agitations (You can also do this mixing in a separate tube). After 30 minutes, shake the slide to remove the reagent.
4. Wash with distilled water.
5. Cover with 10 drops of “C-Methanyl Yellow Solution” and shake the slide after 1 minute to remove the reagent.
6. Wash with distilled water.
7. Dehydrate, clean, cover with synthetic resin and examine under a microscope.
