Purpose
It is used in the examination of silver-retaining reticular fibers in connective and supporting tissues by light microscopy. It is specially produced to be used in paraffin block tissue sections or cytological preparations that are fixed in formalin and routine tissue follow-up.
Ingredients
A- Potassium Permanganate Solution: Potassium permanganate
B- Acid Activator Solution: Sulfuric acid 96%
C- Oxalic Acid Solution: Oxalic acid
D- Ferric Ammonium Sulfate Solution: Ferric ammonium sulfate
E- Silver Nitrate Solution: Silver nitrate
F- Potassium Hydroxide Solution: Potassium hydroxide
G- Ammonium Hydroxide Solution: Ammonium hydroxide 28%
H- Formalin Solution: Formaldehyde 37%
I- Sodium Thiosulfate Solution: Sodium thiosulfate pentahydrate
Working Principle
Silvering reticulum staining is based on the principle of selective staining of reticular fibers by silver ions. In this method, silver ions are reduced by reticular fibers and precipitate as metallic silver, which makes these fibers appear black or dark brown under the microscope.
Warnings, Precautions, Recommendations
Caution. Danger. Harmful if swallowed, in contact with skin or if inhaled. Causes severe skin burns and eye damage. May cause respiratory tract irritation. May cause cancer. Do not breathe dust/fume/gas/mist/vapors/spray. May be corrosive to metals. Very toxic to aquatic life with long lasting effects. Causes serious eye irritation. Causes skin irritation. Avoid release to the environment. Wear protective gloves/protective clothing/eye protection/face protection. IF IN EYES: Rinse cautiously with water for several minutes. Remove contact lenses, if present and easy to do. Continue rinsing. Immediately call the NATIONAL POISON CONSULTATION CENTER ON 114 or a doctor/physician. IF INHALED: Remove victim to fresh air and keep in a position comfortable for breathing. IF SWALLOWED: Rinse mouth. DO NOT induce vomiting. IF ON SKIN: Wash with plenty of soap and water. If skin irritation occurs: Get medical advice/attention. Remove contaminated clothing and wash before reuse. Collect spillage. Keep only in original container.
Application
1. Rinse the preparation with distilled water.
2. Drop 5 drops of 'A- Potassium Permanganate Solution' and 1 drop of 'B- Acid Activator Solution' in order. Wait for 5 minutes.
3. Wash in distilled water.
4. Drop 10 drops of 'C- Oxalic Acid Solution'. Wait for 3 minutes.
5. Wash twice with distilled water.
6. Drop 10 drops of 'D- Ferric Ammonium Sulphate Solution'. Wait for 3 minutes.
7. Wash twice with distilled water.
8. Drop 4 drops of 'E- Silver Nitrate Solution' and 1 drop of 'F- Potassium Hydroxide Solution' to the preparation in order. Drop 2 drops of 'G- Ammonium Hydroxide Solution' on it. Mix with slow oscillations until turbidity disappears. Leave for 3 minutes.
9. Wash in distilled water.
10. Drop 10 drops of ‘H- Formalin Solution’. Leave for 5 minutes.
11. Wash twice with distilled water.
12. Drop 10 drops of ‘I- Sodium thiosulphate Solution’. Leave for 5 minutes.
13. Wash with tap water for 5 minutes.
14. Dehydrate with graded alcohols.
15. Clean and cover with Covering Medium.
