Purpose
Designed for general laboratory use. For the Ziehl Neelsen acid-fast staining method. Divides microorganisms according to whether they are acid-resistant or not.
Ingredients
Fuchsin, phenol, ethanol.
Working Principle
Acid-resistant organisms such as Mycobacterium tuberculosis contain large amounts of lipid substances called mycolic acids in their cell walls. These acids are resistant to staining by ordinary methods such as Gram staining.
The reagents used in Ziehl-Neelsen staining are carbol fuchsin, acid alcohol and methylene blue. Acid-resistant bacilli (ARB) appear bright red after staining.
Warnings, Precautions, Recommendations
Hazard . Highly flammable liquid and vapor. Harmful if swallowed. Causes severe skin burns and eye damage. Suspected of causing genetic damage. Keep away from heat/sparks/flame/hot surfaces - Do not smoke. Do not breathe dust/fume/gas/mist/vapor/spray. Wear protective gloves/protective clothing/eye protection/face protection. IF INGESTED: If you feel unwell, call NATIONAL POISON ADVICE CENTER PHONE 114 or a doctor/physician. IF IN CONTACT WITH SKIN (or hair): Immediately remove all contaminated clothing. Rinse skin with water/shower.
IN CASE OF CONTACT WITH EYES: Rinse carefully with water for several minutes. Remove contact lenses if they are worn and easy to do so. Continue rinsing
Application
1. The smear is stained with Carbol Fuchsin Stain (GBL-2423) by heating from the bottom for 5 minutes.
2. Cooled. Wash with distilled water for 5-10 seconds.
3. Cover the smear with Acid Alcohol (GBL-2426) for 5-30 seconds.
4. Gently washed very briefly with distilled water.
5. The smear is stained with Methylene Blue Alkali Stain, Löffler (GBL-2420) for 1-2 minutes.
6. Wash gently with distilled water. Air-dried and analysed.
