Purpose
The Warthin-Starry silver spirochetes staining technique is a special histological staining method used to detect spirochetes (twisted, spiral-shaped bacteria).
Ingredients
A) Silver Nitrate Solution 2%: Silver nitrate, Citric acid
B) Silver Nitrate Solution 1%: Silver nitrate, Citric acid
C) Gelatin Solution 5%: Gelatin, Citric acid
D) Hydroquinone Solution 0.15%: Hydroquinone, Citric acid
Working Principle
The working principle of the Warthin-Starry staining technique is based on the reaction of silver salts with certain components in the bacterial cell wall, creating a dark black or brown color of the silvered bacterial cells.

Application
1. Before starting to stain, the tissue section on the slide should be washed with distilled water three times and the paraffin should be removed.
2. Dip the slides in Silver Nitrate Solution 1% (GBL- 5064-B). Leave the solution in the microwave for 45-60 seconds
3. until it boils. Remove the slide from the solution. Leave it at room temperature for 1 minute.
Preparation of the Development Solution:
1.5 ml Silver Nitrate Solution 2%
3.75 ml Gelatin Solution 5%
2 ml Hydroquinone Solution 0.15%
4. Mix the solutions thoroughly in the order indicated. It should be prepared before use.
NOTE-1: The glass containers should be warm before mixing the solutions. For this, it is necessary to preheat them in the microwave for 45 seconds.
5. Dip the slides in the development solution. Keep them in the microwave for 45-60 seconds. The solution will begin to boil and brown streaks will appear.
6. Rinse quickly with hot tap water.
7. Rinse with distilled water.
8. Dip in 95% ethyl alcohol.
9. Dip in absolute alcohol.
10. Clean with xylene.
11. Cover with synthetic resin and examine under a microscope.
