Purpose Designed for general laboratory use. Designed for Fournier Kinyoun acid-fast staining method at room temperature.
Ingredients Fuchsin, ethanol, propanol, glycerol, dimethyl sulphoxide, phenol.
Working Principle Mycobacterium, nocardia and corynebacterium do not have taychoic acid in their walls. About 35% of the wall is composed of lipids and especially mycolic acid. It is hypothesised that this feature of the wall increases the resistance of the bacterial wall against acid and alcohol. They do not easily release the dye they have taken; that is, they retain the fuchsin dye. Since the wall of bacilli is resistant to acidic alcohol, the dye remains in the bacilli.
Warnings, Precautions, Recommendations Hazard Highly flammable liquid and vapor. Harmful if swallowed. Causes severe skin burns and eye damage. Suspected of causing genetic damage. May cause cancer. Keep away from heat/sparks/flame/hot surfaces - No smoking. Use only outdoors or in a well-ventilated area. Wear protective gloves/protective clothing/eye protection/face protection. IF INGESTED: If you feel unwell, call NATIONAL POISON ADVICE CENTER PHONE 114 or a doctor/physician. IF IN CONTACT WITH SKIN (or hair): Immediately remove all contaminated clothing. Rinse skin with water/shower. In case of exposure or interaction IF: Get medical advice/attention.
Application 1)Cover the smear with Carbol fuchsin solution FK (GBL-2456). After 3 minutes, shake the slide to remove the reagent.
2)Wash with distilled water for 15 seconds.
3)Wash the smear with Acid alcohol solution (GBL-2426). After 5 seconds, shake the slide to remove the reagent.
4)Wash with distilled water for 15 seconds.
5)Cover the smear with Methylene blue solution (GBL-2420). After 15 seconds, shake the slide to remove the reagent.
6)Wash with distilled water for 15 seconds
7)Air dry, preferably on a heated plate. Examine under a microscope.
