Purpose It is used in the examination and differentiation of protozoan cysts and trophozoites by light microscopy.
Ingredients A) Schaudinn Fixative: Ethanol 96%, Glacial acetic acid, Mercury (II) chloride.
B) Alcoholic Solution: Ethanol 96%.
C) Iodine Solution: Potassium iodide, Iodine
D) Wheatley Trichrome Stain: Fast fuchsin G, Phosphotungstic acid, Light green SF, Glacial acetic acid.
E) Acetic Ethanol: Glacial acetic acid, Ethanol 96%.
Working Principle It is a rapid and simple procedure that allows uniform staining of intestinal protozoa, human cells, yeast and artificial materials. Wheatley trichrome stain contains two dyes. Chromotrope 2R stains nuclear chromatin, chromatoid bodies, karyosomes, parasite eggs and larvae, bacteria and digested erythrocytes red to violet-red. Pale green dyes stain the cytoplasm of preserved cysts, trophozoites and cellular components blue-green.
Warnings, Precautions, Recommendations Danger. Highly flammable liquid and vapour. Causes severe skin burns and eye damage. Keep away from heat/sparks/flames/hot surfaces. - No smoking. Wear protective gloves/protective clothing/eye protection/face protection. Keep container tightly closed. Dispose of contents/container in accordance with national regulations. IF ON SKIN (or hair): Remove/take off immediately all contaminated clothing. Dry skin with water/shower. IF IN EYES: Rinse cautiously with water for several minutes. Remove contact lenses, if present and easy to do. Continue rinsing. Store in a well-ventilated area.
Application 1. Before the smear dries (immediately) it is dipped into A- Schaudinn Fixative. The smear is fixed by keeping it in this way for at least 30 minutes. If the situation requires, it can be left in the fixative overnight.
2. The preparation is kept in B- Alcoholic Solution for 5 minutes.
3. It is kept in Iodine Working Solution for 2 minutes.
4. It is kept in B- Alcoholic Solution for 3 minutes.
5. It is kept in B- Alcoholic Solution again for 3 minutes.
6. The slides fixed with Schaudinn Fixative are kept in D- Wheatley Trichrome Stain for 10 minutes.
7. The excess dye of the slides is drained by placing them upright on paper towels.
8. The slides are dipped in E- Acetic Ethanol for 2-3 seconds.
9. After touching the paper towel, the slides are rinsed in 100% alcohol.
10. They are kept in 100% alcohol for 5 minutes.
11. They are kept in 100% alcohol for 5 minutes again.
12. They are kept in toluene or xylene for 5 minutes.
13. They are kept in toluene or xylene for 5 minutes again.
14. Entellan or another coating solution is dropped on the slides and they are covered with coverslips.
15. A drop of immersion oil (GBL-2305) is dropped on the stained slides and they are examined with a x100 objective under a light microscope.
