Purpose It is used to visualize specific structures and components during the examination of connective and supportive tissue or cell samples.
Ingredients A) Weigert Hematoxylin-A Solution: Methanol, Ethanol, 96%, Hematoxylin
B) Weigert Hematoxylin-B Solution: 37% Hydrochloric acid
C) VanGieson Picro-Acid-Fucsin Solution: Picric acid, Fucsin
Working Principle Among the active ingredients in the kit, hematoxylin is attracted to the nucleus, picric acid to the cytoplasm and acid fuchsin to collagen. It is one of the oldest methods used in the study of animal connective tissues. It was first published by vanGieson in 1889. It has undergone many modifications until today. The main modification was made by Mallory in 1936. This set is based on this modification.
Warnings, Precautions, Recommendations Hazard Highly flammable liquid and vapor. Harmful if swallowed. Causes damage to organs. Keep away from heat/sparks/flame/hot surfaces. Wear protective gloves/protective clothing/eye protection/face protection. IN CONTACT WITH SKIN (or hair): Immediately remove all contaminated clothing. Dry skin with water/shower. In case of exposure: Call the NATIONAL POISON ADVICE CENTER, TELEPHONE 114 or a doctor/physician.
Application 1. Add 5 drops of “A - Weigert hematoxylin solution A” and 5 drops of “B - Weigert hematoxylin solution B” to the preparation. Mix the two dye solutions by blowing and small oscillations. After 10 minutes, remove the reagent by shaking the slide.
2. Wash with distilled water using the overflow method for 5 minutes.
3. Add 10 drops of “C- vanGieson picro-acid-fucsin solution” to the preparation. After 10 minutes remove the reagent by shaking the slide.
4. Wash with distilled water for 3 seconds.
5. Quickly dehydrate with alcohol for 1 minute, clean, cover with synthetic resin and examine under a microscope. Examine under a microscope.
