Purpose It is used to examine the peroxidase activities of leukocytes.
Ingredients A) Benzidine solution: Ethanol 96%, Benzidine, Sodium nitroprusside
B) Hydrogen peroxide solution: Hydrogen peroxide, Acetanilide, Phosphate buffers
C) Hematoxylin Solution Gill-3: Hematoxylin, Sodium iodate, Ethylene glycol, Acetic acid
Working Principle Peroxidase Leukocyte Staining Set uses a suitable substrate to determine the activity of peroxidase enzymes. Peroxidase enzymes hydrolyze the substrate to form a colored product. This colored product allows peroxidase positive cells to be easily detected under the microscope.
Warnings, Precautions, Recommendations Caution. Danger. Harmful if swallowed. Causes serious eye irritation. Causes skin irritation. Highly flammable liquid and vapour. Do not breathe dust/fume/gas/mist/vapours/spray. May cause damage to organs through prolonged or repeated exposure. Wear protective gloves/protective clothing/eye protection/face protection. Causes serious eye irritation. Keep away from heat/sparks/flames/hot surfaces. - No smoking. Keep container tightly closed. IF ON SKIN (or hair): Remove/take off immediately all contaminated clothing. Dry skin with water/shower. IF IN EYES: Rinse cautiously with water for several minutes. Remove contact lenses, if worn and easy to do. Continue rinsing.
Application 1. Cover the smear with 10 drops of A-Benzidine Solution. Wait 1 minute. Do not pour the reagent.
2. Add approximately 1-2 loopfuls (10-20 microliters) of B-Hydrogen Peroxide Solution to the first reagent. Mix by blowing and small agitations. After 1 minute, shake the slide to remove the reagent.
3. Wash in distilled water for 1 minute.
4. Cover the smear with 10 drops of C-Hematoxylin Solution Gill-3. After 1.5 minutes, shake the slide to remove the reagent.
5. Wash in distilled water for 30 seconds.
6. Dry in air. Examine under a microscope with an immersion lens.
