Purpose It is used to stain mast cells in the connective tissue and examine them using light microscopy.
Ingredients A) Toluidine Blue Stock Dye: Toluidine blue, 96% Ethanol
B) Sodium Chloride Solution: Sodium chloride
Working Principle Toluidine blue is the unique binding of the dye substance by interacting with the acidic components in the sample. In particular, toluidine blue has metachromasia properties, that is, it has the ability to stain different components in different colors. The chemical structure of the cells, tissue structures or other components in the sample interacts with toluidine blue and allows them to be stained in a certain color.
In this way, toluidine blue staining is used to highlight certain structures in histological or cytological samples and to better visualize them under the microscope. In particular, this staining method is widely used in highlighting connective tissue matrix, mast cells and some other cell types.
Warnings, Precautions, Recommendations
Application Preparation of Toluidine Blue Working Solution:
A- Toluidine Blue Stock Dye 5 mL
B- Sodium Chloride Solution 1% 45 ml
1. In a clean container, mix the two solutions in the ratios indicated above. (1:9 ratio)
2. The final pH should be less than approximately 2.5. The contrast quality of the staining decreases at pH values ​​higher than 2.5.
3. This solution should be prepared fresh and should be discarded after use.
4. Fixation: With 10% Formalin fixative.
5. Staining Method: Covering method
STAINING:
1. Rinse the preparation with distilled water.
2. Cover the preparation with the “Toluidine Blue Working Solution” as described above. Wait 2-3 minutes.
3. Rinse the preparation with distilled water 3 times.
4. Dehydrate by quickly rinsing with 95% and 100% ethyl alcohol (2 times). (After 10 dips for each preparation, the dye stains will disappear quickly.)
5. Clean with xylene, cover with xylene-based synthetic resin.
6. Examine under a microscope.
