Purpose Bouin's fixative is a non-coagulant yellow dye fixative. Due to its picric acid component, it causes shrinkage in tissues when used as a preservative. It is especially used in ureteroscopic biopsies and preserves morphological and nuclear information in tissues after fixation.
Since Bouin's Fixative preserves cell nuclei and chromosomes well, it is especially preferred for tissues in which chromosomes will be observed during mitosis and meiosis.
Ingredients Picric acid, formaldehyde, glacial acetic acid.
Working Principle Bouin's Fixative is commonly used in histology. Bouin's Fixative is particularly useful for fixation of gastrointestinal tract biopsies because this fixative provides more viable and better nuclear staining than 10% neutral buffered formalin. It is a good fixative when tissue structure with soft and delicate tissue needs to be preserved. The acetic acid in this fixative breaks down red blood cells and dissolves small deposits of iron and calcium in the tissue. The effects of the three chemicals in Bouin's solution balance each other. Formalin causes the cytoplasm to become basophilic, but this effect is counterbalanced by the effect of picric acid. The tissue-hardening effect of formalin is offset by the soft tissue fixation of picric and acetic acids. The tissue-inflating effect of acetic acid is counterbalanced by the tissue-shrinking effect of picric acid.
