Purpose Wright-Giemsa Stain is a staining solution used for cellular examination and diagnostic purposes. This staining solution is frequently used in hematologic examinations of blood preparations and other body fluids. In particular, it is used to clarify the morphology and structure of cells on preparations such as peripheral blood smears and bone marrow smears.

Typically, Wright-Giemsa Stain stains the nucleus (nucleus) of cells blue, while staining the cytoplasm (intracellular fluid) and cell organelles pink or red. This combination of colors highlights the details and structures of cells, making it easier to identify pathological conditions or diseases.

Warnings, Precautions, Recommendations Danger Toxic Highly flammable liquid and vapor. Toxic by ingestion, skin contact or inhalation. Causes damage to organs. Keep away from heat/sparks/flame/hot surfaces - No smoking. Wear protective gloves/protective clothing/eye protection/face protection. IF IN CONTACT WITH SKIN (or hair): Immediately remove all contaminated clothing. Rinse skin with water/shower. In case of exposure: Call NATIONAL POISON ADVICE CENTER PHONE 114 or doctor/physician. Store in a well ventilated area. Keep container tightly closed. Store under lock and key.
Application Implementation of the staining method
A)Preparation of Dye Solution (Dilute Giemsa Solution)
Add 5 ml Wright-Giemsa Solution (GBL_2300) 5 ml Giemsa Buffer pH 6.8 (GBL-2307) 90 ml distilled water to the staining container, mix.
B) Preparation of buffered water working solution
Mix 5 ml Giemsa Buffer pH 6.8 (GBL-2307) and 95 ml distilled water.
Staining with Wright-Giemsa Solution
1-)Cover the preparation with methanol. Fix for 5 minutes.
2-) Immerse/ cover the preparation in the prepared staining solution. After 15-30 minutes, remove the slide and shake to remove the reagent.
3-)Wash with distilled water. Wash with buffered water working solution. Dry it.
4-)Examine under microscope.
Staining with May-Grünwald solution and Wright-Giemsa solution
C)For staining on the staining rack;
1-)Cover the preparation with 1 ml of May-Grünwald solution. Stain for 3 minutes.
2-)Add 1 ml of buffered water working solution, mix and stain for 5 minutes.
3-)Cover with dilute Wright-Giemsa solution and stain for 15-20 minutes.
4-)Rinse with buffered water working solution. Let dry. Examine under microscope.
D) For staining in staining waterfall;
1-)Stain with May-Grünwald solution for 3-5 minutes.
2-)Stain with diluted Wright-Giemsa solution for 15-20 minutes.
3-)Rinse 2 times for 1 minute with buffered water working solution. Dry. Examine under microscope.
