Purpose It is used to demonstrate glycols in leukocytes by light microscopy.
Ingredients A) Periodic acid solution: Periodic acid
B) Schiff reagent: Pararosaniline, Sodium metabisulfite, Hydrochloric acid, Activated medical charcoal
C) Hematoxylin solution Gill-3: Ethylene glycol, Hematoxylin, Sodium iodate, Acetic acid, Aluminum sulfate
Working Principle Periodic acid selectively oxidizes 1,2-glycolic, primary amine, secondary amine and hydroxyketonic groups. High molecular weight carbohydrates carry these groups. As a result of oxidation, the ends of some carbon atoms in the structure become free. It holds the leukofuksin dye (sulfurized fuchsin dye) and prevents it from dissolving in water. The parts where the dye is held are seen in magenta red color.
Warnings, Precautions, Recommendations Toxic. Harmful. Irritating to eyes and skin. Harmful if swallowed or inhaled. Prolonged exposure may cause shortness of breath, mucosal irritation, coughing. May cause cancer. IF exposed or exposed: Get medical advice/attention. Store locked up. In eyes: May have corrosive effects. In case of contact with eyes, rinse immediately with plenty of water and seek medical advice. On skin: May have corrosive effects. Remove contaminated clothing and shoes immediately. Wash with soap and plenty of water. Consult a physician. If swallowed: Do not induce vomiting. Never give anything by mouth to an unconscious person. Rinse mouth with water. Consult a physician.
Application 1. Cover the tissue section with 10 drops of “A-Periodic acid solution”. After 5 minutes, shake the slide to remove the reagent.
2. Wash with distilled water for 10 seconds.
3. Cover with 10 drops of “B-Schiff reagent”. After 15 minutes, shake the slide to remove the reagent.
4. Wash with distilled water for 5 minutes.
5. Cover with 10 drops of “C-Hematoxylin solution Gill-3”. After 1.5 minutes, shake the slide to remove the reagent.
6. Wash with distilled water for 30 seconds.
7. Air dry and examine under a microscope with immersion oil. You can cover the smear if you wish.
