Purpose |
Vet-Quick Staining Kit is designed as a staining kit used in veterinary medicine. This kit is used for fast and efficient staining of veterinary specimens. It is commonly used for staining various samples such as tissue sections, cell fluids or cytological specimens. |
Ingredients |
A) Methanol Fixative: Dark Green FCF, Formaldehyde, Methanol.
B) Buffered Xanthene Solution: Eosin Y, Eosin B, Phloxin B, Phosphate buffers, Tiyoüre, Sodyum azid.
C) Buffered Thiazine Solution: Phosphate buffers, Methylene blue, Azide A, Sodium azide. |
Working Principle |
Methanol is used for fixation of cell and tissue samples. Fixation preserves cell structures and facilitates the examination of morphological details under a microscope. Xanthene stains are often used to stain cell cytoplasm and other acidic structures. Xanthene dyes interact with and color cell and tissue components. Thiazine dyes are commonly used to stain the cell nucleus and other basic structures. Thiazine dyes interact with, and color negatively charged components such as nucleic acids. The buffered solution maintains the pH balance of the dye and ensures optimal staining conditions. |
Warnings, Precautions, Recommendations |
Danger. Causes allergic skin reactions. Highly flammable liquid and vapor. Toxic if swallowed, in contact with skin, or if inhaled. Suspected of causing cancer. Causes damage to organs. Keep away from heat/sparks/open flames/hot surfaces. – No smoking. Wear protective gloves/protective clothing/eye protection/face protection. Use personal protective equipment. IF SWALLOWED: Call the NATIONAL POISON CONTROL CENTER at 114 or doctor/physician. IF ON SKIN: Wash with plenty of soap and water. IF INHALED: Remove the person to fresh air and keep comfortable for breathing. If exposed or concerned: Get medical advice/attention. |
Application
|
1. Immerse the air-dried smear in a beaker of “A-Methanol fixative”.
2. Remove the reagent by dipping into the dye 1 time every second (10 times in 10 seconds), shaking the slide after 10 seconds.
3. Immerse the preparation in a beaker of “B- Buffered xanthene solution”.
4. Immerse the preparation in the dye 1 time every second (6 times in 6 seconds), after 6 seconds shake the slide to remove the reagent.
5. Immerse the preparation in a beaker of “C-buffered thiazine solution”.
6. Remove the reagent by dipping into the dye 1 time per second (6 times in 6 seconds), shaking the slide after 6 seconds.
7. Wash with distilled water for 1 minute.
8. Air dry. Examine under a microscope.
