Purpose It is used to stain the transparent and unstained capsules of microorganisms using the negative staining method.
Ingredients Nigrosin, Formaldehyde
Working Principle The negative staining method allows the visualization of the usually transparent and non-staining capsules of many organisms, most notably Cryptococcus neoformans. Nigrosin consists of a suspension of fine carbon particles. These form a dark background against which the capsules are clearly visible as a result of the displacement of the carbon particles.
Warnings, Precautions, Recommendations May cause sensitization by skin contact. Wear protective gloves/protective clothing/eye protection/face protection.
Application 1.Place 1 drop of Nigrosin Stain on a clean microscope slide, adjacent to the frosted edge.
2.Using a flamed loop and sterile technique, mix some microorganisms with the Nigrosin Stain on the slide. Make sure there are no large clumps of microorganisms.
3.Place the end of another clean microscope slide at an angle to the end of the slide containing the microorganism and spread the stain by pulling the slide.
4.Let the film dry.
5.Observe the slide under the microscope using appropriate microscopy technique.
