Purpose It is often used for differentiation (decolorisation) during regressive staining.
Ingredients Ethyl alcohol, Methanol and Hydrochloric Acid.
Working Principle Acid alcohol is a differentiation reagent. It is often used in regressive staining methods. It provides an excellent separation between nuclear and non-nuclear structures. Differentiation occurs when acidic alcohol removes the dyes from the cytoplasm, while the nucleus remains stained. This is because the nuclear dye binds more strongly to the nucleus than to the cytoplasm.
