Purpose
It is used in microbiology and clinical diagnostics, especially for the detection of acid-resistant bacteria (ARB) such as Mycobacterium tuberculosis. The purpose of this solution is to allow such bacteria to accumulate in the cell walls and to be seen brightly under a fluorescence microscope
Ingredients
Auramine o, Rhodamine B, Glycerol, Phenol, Propanol
Working Principle
It is a staining technique used to visualize acid-fast bacilli using fluorescence microscopy. Acid-fast organisms exhibit a reddish yellow fluorescence color. Although the auramine-rodamine stain is not as specific as the Ziehl-Neelsen stain for acid-fast organisms (e.g. Mycobacterium tuberculosis or Nocardia), it gives faster results, so is often used as a screening tool.
Warnings, Precautions, Recommendations
Danger. May cause severe skin burns and eye damage. Suspicion of genetic damage may occur. Use personal protective equipment. Wear personal protective equipment. If swallowed: Rinse mouth. DO NOT attempt to void.
IF IN CONTACT WITH SKIN (or hair): Immediately remove all contaminated clothing. Rinse skin with water/shower. IN CASE OF CONTACT WITH EYES: Rinse carefully with water for several minutes. Remove contact lenses if fitted and easy to do so. Continue rinsing. Keep under lock and key.
Application
1. Cover with “A- Auramine Rhodamine Solution”. Shake off after 15 minutes.
2. Wash with distilled water.
3. Cover with “B- Acidic Bleaching Solution”. Shake off after 1 minute. You can change the reagent many times during this time.
4. Wash with distilled water.
5. Cover with “C- Potassium Permanganate Solution”. After 3 minutes, shake the preparation to remove the reagent.
6. Wash with running distilled water.
7. Examine under a microscope.
