Purpose It is used in the examination of sulfated and carboxylated mucins under a light microscope.
Ingredients A) N,N-dimethyl-meta-phenylenediamine dihydrochloride: N,N-dimethyl-meta-phenylenediamine dihydrochloride
B) N,N-dimethyl-para-phenylenediamine dihydrochloride: N,N-dimethyl-para-phenylenediamine dihydrochloride
C) Ferric chloride solution: Ferric chloride
D) Alcian blue pH 2.5 solution: Alcian blue, glacial acetic acid
E) Neutral red acetic solution: Nuclear fast red, glacial acetic acid
Working Principle
Warnings, Precautions, Recommendations Caution. Toxic. Irritant. Causes serious eye damage. Causes skin irritation. If inhaled: Prolonged exposure may cause breathing difficulties, mucosal irritation, coughing, and headaches. If swallowed: Seek medical attention. Suspected of causing cancer. Use protective gloves/protective clothing/eye protection/face protection. If it comes into contact with the skin: Immediately remove all contaminated clothing. Rinse the skin with water/shower. If it comes into contact with the eyes: Rinse carefully with water for several minutes. If possible, remove contact lenses. Continue rinsing. If exposed or affected: Seek medical attention/care. Store under lock and key.
Application SAMPLES AND PREPARATION
Add one tube each of reagents A and B to a container and dissolve in a quantity of distilled water. Finally, add 1.5 ml of reagent C. Mix and dissolve. Make up to 50 ml with distilled water.
High-concentration iron-containing phenylenediamine solution:
Reagents 50 ml container
Distilled water 50 ml
A- N,N-dimethyl-meta-phenylenediamine dihydrochloride 120 mg 1 tube
B- N,N-dimethyl-para-phenylenediamine dihydrochloride 20 mg 1 tube
C- Ferric chloride solution 1.5 ml

STAINING METHOD
1. Place the preparation in the “High-concentration iron-containing phenylenediamine solution” container at room temperature. Close the container. After 18-24 hours, remove the slide and shake to remove the separator.
2. Rinse with distilled water for 1 minute.
3. Cover the preparation with 10 drops of “D- Alcian blue pH: 2.5 solution.” After 5 minutes, shake the slide to remove the separator.
4. Rinse with distilled water for 1 minute.
5. Cover the preparation with 10 drops of “E- Neutral red acetic solution.” After 3 minutes, shake the slide to remove the separator.
6. Wash with distilled water for 1 minute.
7. Dry, clean, cover with synthetic resin, and examine under a microscope.
