Wright eosin methylene blue dye, surfactant, buffers, stabilizers and methanol. |
Working Principle |
Wright Stain is a polychromatic stain consisting of a mixture of Eosin and Methylene blue. Since Wright stain is methanol based, it does not require a fixation step before staining. Methanol fixes the cells to the slide. Eosin Y is an acidic anionic dye, while methylene blue is a basic cationic dye. Ionization occurs when diluted in buffered water. Eosin stains basic components such as hemoglobin and eosinophilic granules. Methylene blue stains acidic cellular components such as nucleic acids and basophilic granules. Neutral components of the cells are stained by both components of the stain, producing variable colors. |
Warnings, Precautions, Recommendations |
Danger. May cause an allergic skin reaction. Highly flammable liquid and vapour. Toxic if swallowed, in contact with skin or if inhaled. Causes damage to organs. Keep away from heat/sparks/flames/hot surfaces. - No smoking. Wear protective gloves/protective clothing/eye protection/face protection. IF ON SKIN (or hair): Remove/Take off immediately all contaminated clothing. Rinse skin with water/shower. IF INHALED: Remove victim to fresh air and keep at rest in a position comfortable for breathing. In case of exposure: Call the NATIONAL POISON CONSULTATION CENTER ON 114 or doctor/physician. Store locked up. |
Application
|
1.Approximately 50 ml of Wright Stain (GBL-2413) is filled into the container where the preparation will be immersed.
2.Deionized water is also filled into another container where the preparation will be immersed.
3.The prepared preparation is first immersed in the dye solution and waited for approximately 1-3 minutes.
4.The preparation removed from the dye solution is immersed in the container filled with deionized water and waited for 45 seconds.
NOTE: Do not move the preparation while it is in water or wash the preparation in running water.
5.Remove the preparation from the deionized water by shaking it gently after the waiting period.
6.Let the preparation dry and examine it under a microscope |
