Purpose Elastic Van Gieson (EVG) is a special staining method used in histology and pathology laboratories. This technique is used to highlight elastic fibers and collagen fibers.
Ingredients A) Hematoxylin solution, Alcoholic: Hematoxylin, 96% Ethanol
B) Ferric chloride solution: Iron (III) chloride
C) Weigert iodine solution: Iodine, Potassium iodide
D) VanGieson solution: Acid fuchsin, Picric acid
Working Principle Elastic dyes bind to elastin proteins, causing elastic fibers to appear dark. This helps to highlight elastic fibers in tissue sections. A mixture of pico acid and acidic fuchsin stains collagen fibers red and other connective tissue components yellow. This differential staining allows collagen and elastic fibers to be easily distinguished from each other. The EVG staining method provides important information in textural and pathological examinations by highlighting elastic and collagen fibers. The dark purple/black appearance of elastic fibers and red appearance of collagen fibers allows these structures to be easily distinguished under the light microscope.
Warnings, Precautions, Recommendations Explosive. Toxic. Irritant. Harmful. Highly flammable liquid and vapour. Causes skin irritation. Causes serious eye irritation. Keep away from heat/sparks/flames/hot surfaces. – No smoking. IF ON SKIN (or hair): Remove/take off immediately all contaminated clothing. Wash with plenty of soap and water. Rinse skin with water/shower. If skin irritation occurs: Get medical advice/attention.
Application 1. Rinse the preparations with distilled water.
2. Stain with Elastic Staining Working Solution for 10 minutes.
3. At the end of the staining period, wash the preparations with distilled water.
4. Differentiate with 2% Ferric Chloride Working Solution.
5. Wash with tap water.
6. Perform the necessary check under the microscope. If the differentiation is high, return to the Elastic Staining Working Solution step.
7. Wash the preparation with 95% ethyl alcohol to remove iodine in the medium.
8. Wash with distilled water.
9. Stain with “D-Van Gieson solution” for 1-3 minutes.
10. Wash with 95% ethyl alcohol.
11. Dehydrate the medium with xylene. Cover.
