Purpose
It is used to stain metachromatic bodies in the bacterial cytoplasm.
Ingredients
A) Neisser-A Dye Solution: Ethanol 96%, Methylene Blue, Acetic Acid
B) Neisser-B Dye Solution: Crystal Violet, Ethanol 96%
C) Neisser-C Dye Solution: Chrysoidin G Dye, Preservative
D) Iodine Solution: Iodine, Potassium Iodide
Working Principle
The Neisser staining method specifically stains the metachromatic granules of bacteria, making these granules visible under the microscope. Neisser A and Neisser B stains interact specifically with metachromatic granules.
Warnings, Precautions, Recommendations
Danger. Causes severe skin burns and eye damage. Wear protective gloves/protective clothing/eye protection/face protection. IF SWALLOWED: Rinse mouth. DO NOT induce vomiting. IF ON SKIN (or hair): Remove/Take off immediately all contaminated clothing. Rinse skin with water/shower. IF IN EYES: Rinse cautiously with water for several minutes. Remove contact lenses, if present and easy to do. Continue rinsing.
Application
1. Mixed stain is prepared from Neisser-A and Neisser-B dye solutions.
2. Spread the mixed stain on the preparation. Leave for 20-30 seconds.
3. Wash with distilled water for 15 seconds.
4. Drop 10 drops of “D-Lugol’s solution” on the preparation. Leave for 5 seconds.
5. Wash with distilled water for 15 seconds.
6. Drop 10 drops of “C-Neisser-C dye solution” on the preparation. Leave for 10 seconds.
7. Wash with distilled water for 15 seconds. Dry with clean paper.
8. Pass tissue samples through graded alcohols quickly. Clean with xylene, cover with coverslip. Examine under a microscope.
9. Examine smears under a microscope with an immersion lens.
