Purpose It is used to examine DNA and RNA in tissue sections by light microscopy.
Ingredients A) Acidic alcohol: 96% Ethanol, 37% Hydrochloric acid
B) Alcyan blue solution: Alcyan blue, Acetic acid
C) Alcoholic buffer solution: Acetate buffer, 96% Ethanol, Acetic acid
D) Methyl green pyronine solution: Methyl Green, Pyronin Y, Glycerol, Acetic acid
Working Principle Methyl Green Pyronine staining kit is a staining method used to distinguish DNA and RNA in histological examinations by staining them in different colors. This method specifically stains DNA in the cell nucleus and RNA in the cytoplasm so that these two types of nucleic acids appear in different colors. DNA is found in the nucleus (nucleus). RNA is found in the nucleolus and cytoplasm. DNA is stained with methyl green and RNA with pyronine. Acidic Alcohol removes excess stain. Alcyan Blue Solution stains mucus and acidic mucopolysaccharides. Alcoholic Buffer Solution stabilizes the structure of the cells and provides the appropriate pH for staining.
Warnings, Precautions, Recommendations Caution Danger Causes skin irritation. Serious eye irritation. Suspected of causing cancer. Suspected of causing damage to the unborn child or reproductive harm. Causes organ damage through prolonged or repeated exposure. Flammable liquid and vapor. Keep away from heat/sparks/flame/hot surfaces. - Do not smoke. IF IN CONTACT WITH SKIN (or hair): Immediately remove all contaminated clothing. Wash with plenty of soap and water. Rinse skin with water/shower. In case of skin irritation: Get medical advice/attention.
Application 1. Cover the preparation with 10 drops of “A-Acid alcohol”. After 5 minutes, shake the slide to remove the reagent.
2. Dry with absorbent paper.
3. Cover the preparation with 10 drops of “B- Alcyan blue solution”. After 15 minutes, shake the slide to remove the reagent.
4. Dry with absorbent paper.
5. Cover the preparation with 10 drops of “C- Alcoholic buffer solution”. After 10 minutes, shake the slide to remove the reagent
5. Wash with distilled water for 10 minutes.
6. Cover the preparation with 10 drops of “D- Methyl green pyronine solution”. After 7 minutes, shake the slide to remove the reagent.
7. Wash with distilled water for 15 seconds.
8. Dry with absorbent paper.
9.Air dry for no more than 10 minutes.
10. Clean with xylene. Cover. Examine under a microscope.
