Purpose |
It is used to detect acid-resistant bacteria by staining. |
Ingredients |
A) Carbol Fuchsin Solution ZN: Fuchsin, Phenol, Ethanol, Ro Water.
B) Acid Alcohol Solution: Methanol, Ethanol, Hydrochloric Acid Solution.
C) Methylene Blue Solution: Methylene Blue, Ethanol, Tween-80, Phosphate Buffers. |
Working Principle |
Mycobacterium, nocardia and corynebacterium do not have taychoic acid in their walls. About 35% of the wall is composed of lipids and especially mycolic acid. It is hypothesised that this feature of the wall increases the resistance of the bacterial wall against acid and alcohol. They do not easily release the stain they have taken up, i.e. they retain the Fuchsin stain. Since the wall of the bacilli is resistant to acidic alcohol, the dye remains in the bacilli. |
Warnings, Precautions, Recommendations |
Hazard Highly flammable liquid and vapor. Harmful if swallowed. Causes severe skin burns/irritation and eye damage/irritation. Causes damage to organs. Suspected of causing genetic damage. Keep away from heat/sparks/flame/hot surfaces. Do not smoke. Do not breathe dust/fume/gas/mist/vapor/spray. Wear protective gloves/protective clothing/eye protection/face protection. IF INGESTED: If you feel unwell, call NATIONAL POISON ADVICE CENTER PHONE 114 or a doctor/physician. IF IN CONTACT WITH SKIN (or hair): Immediately remove all contaminated clothing. Rinse skin with water/shower. IN CASE OF CONTACT WITH EYES: Rinse carefully with water for several minutes. Remove contact lenses if fitted and easy to do so. Continue rinsing. If eye irritation persists: Get medical advice/attention. Store in a well-ventilated area. Keep cold. |
Application
|
1.Cover the preparation with "A-Carbol Fuchsin Solution ZN".
2.Heat from the bottom for not less than 5 minutes.
3.Wash with distilled water for 15 seconds.
4.Cover the preparation with "B-Acid Alcohol Solution". After 15 seconds, shake the slide to remove the reagent.
5.Wash with distilled water for 15 seconds.
6.Cover the preparation with "C-Methylene Blue Solution". After 30 seconds, shake the slide to remove the reagent.
7.Wash with distilled water for 15 seconds.
8.Air dry, preferably on a heated plate. Examine under a microscope. |
