Purpose It is used to examine naphthol AS-D chloroacetate esterase activity in leukocytes.
Ingredients A) Citrate solution pH: 5.4: Citric Acid, Sodium Chloride, Chloroform
B) FRV red dye: Fast Red Violed LB salt
C) Sodium nitrite solution: Sodium nitrite
D) Trismalate solution pH: 6.3: Tris buffer, Tween-20
E) Naphthol AS-D chloroacetate solution: Naphthol AS-D chloroacetate
F) Hematoxylin solution Gill-3: Hematoxylin, Sodium iodate, Acetic acid
Working Principle The Specific Esterase Leukocyte Staining Kit uses a substrate to determine the presence and activity of esterase enzymes. Esterases hydrolyze the substrate to form a colored product. This allows specific types of leukocytes to be easily identified under the microscope.
Warnings, Precautions, Recommendations Caution. Harmful if swallowed, in contact with skin or if inhaled. Causes skin irritation. Causes serious eye irritation. May cause organ damage through prolonged or repeated exposure.
Suspected of causing cancer. Avoid breathing dust/fume/gas/mist/vapors/spray. Wear protective gloves/protective clothing/eye protection/face protection. IF IN EYES: Rinse cautiously with water for several minutes. Remove contact lenses, if present and easy to do. Continue rinsing. If you feel unwell, call a POISON CENTER or doctor/physician. IF SWALLOWED: If you feel unwell, call the NATIONAL POISON CONSULTATION CENTER ON 114 or a doctor/physician. Rinse mouth.
Application 1. Immerse the preparation in the container containing the “Fixative Solution”. After 30 seconds, shake off the slide and remove the reagent.
2. Wash with distilled water for 1 minute.
3. Immerse the preparation in the container containing the incubation solution. Incubate at 37 degrees C and protected from light. After 15 minutes, remove the slide and shake off the reagent.
4. Wash with distilled water for 2 minutes.
5. Stain with F- Hematoxylin solution Gill-3 for 2 minutes. At the end of the period, shake off the slide and remove the reagent.
6. Wash with distilled water for 15 seconds.
7. Dry in air. Examine under a microscope. If desired, use only aqueous covering medium.
