Purpose It is used to stain cells found in urine sediment.
Ingredients Crystal violet, Ammonium oxalate, Safranin, Ethanol, Stabilizers.
Working Principle With the help of dyes in the product content, cells in the urine sample are stained and manually detected under a microscope.
Warnings, Precautions, Recommendations
Application 1. Take a centrifuge tube and put 15 ml of urine sample in it.
2. Centrifuge at 1,500 rpm for 2-3 minutes.
3. Empty the top until 0.3 ml of urine remains at the bottom.
4. Try to obtain a homogeneous mixture by mixing the remaining urine sample.
5. Add 1 drop of “Urine Sediment stain”.
6. Mix and wait for 2-3 minutes.
7. Take 2 drops of the stained urine sample and place it on the slide.
8. Examine under the microscope by placing a coverslip on it so that no air bubbles are formed.
9. Scan the area with x100 magnification. Re-examine suspicious areas with x400 magnification. Non-bright light should be used to better examine cellular elements. After scanning 20 areas with x400 magnification, record the average of these.
10. Polymorphic nucleated leukocytes are very common in stained preparations. Eosinophils and lymphocytes may also be found, although not in large numbers.
Theoretical Results
Neutrophils Cells that are pale (their cytoplasm does not take up any stain) or have fragmented and unstained nuclei or are dark in color.

Cytoplasm Dusty rose color or colorless
Granules in the cytoplasm Dark Violet
Erythrocytes Light greenish-yellow
