Purpose |
It is the counting stain of the Gram staining method. With this staining procedure, microorganisms are separated as gram positive and gram negative. |

Working Principle |
Gram Positive/Negative status is a feature of the cell wall (peptidoglycan) structure of bacteria. The peptidoglycan structure of Gram Negative bacteria is thinner between the outer membrane and inner membrane than Gram Positive bacteria. In Gram positive bacteria, the thick peptidoglycan is capable of retaining crystal violet. Lugol ensures the retention of the dye. Cell wall permeability of Gram positive bacteria is less. Therefore, they do not release dyes during decolourisation with denatured alcohol. In Gram-negative bacteria, alcohol dissolves the lipids in the outer membrane, the pores open and the crystal violet disperses. Gram-negative bacteria are then stained with fuchsin added to the medium.

Application
|
1)If the smear is too acidic, cover with Sodium Hydrogen Carbonate Solution for 15-30 seconds.
2)Cover the smear with Crystal Violet Stain (GBL-2434) for 1-2 minutes.
3)Wash with distilled water.
4)Cover the smear with Gram Lugol's Solution (GBL-2435) for 1 minute.
5)Washed with distilled water.
6)Cover the smear with Denatured Alcohol (GBL-2891) for 5-15 seconds.
7)Washed with distilled water.
8)Cover the smear with Gram Fuchsin Aqueous Stain (GBL-2424) for 1 minute.
9)Gently washed with distilled water. Air dried and analysed. |
