A) Periodic Acid Solution: Periodic acid.
B) Schiff Reagent McManus: Pararosaniline, sodium metabisulfite, hydrochloric acid, activated charcoal.
C) Sodium Metabisulfite Solution: Sodium metabisulfite.
D) Detection Solution: Sodium metabisulfite, hydrochloric acid.
E) Mayer Hemalum Solution: 96% Ethanol, hematoxylin monohydrate, ammonium aluminum sulfate, sodium iodate, glycerol, triton X-100, citric acid monohydrate. |

| Application
|
1. Cover the tissue section with 10 drops of “A-Periodic Acid Solution”. After 10 minutes, shake the slide to remove the reagent.
2. Wash with distilled water for 2 minutes.
3. Cover with 10 drops of “B-Schiff Reagent McManus”. After 20 minutes, shake the slide to remove the reagent.
4. Wash with distilled water for 2 minutes.
5. Cover with 10 drops of “C-Sodium Metabisulfite Solution”. After 2 minutes, shake the slide to remove the reagent.
6. Cover with 10 drops of “D-Detection Solution” without rinsing with water. After 2 minutes, shake the slide to remove the reagent.
7. Wash with distilled water for 2 minutes.
8. Cover with 10 drops of “E-Mayer Hemalum Solution”. After 3 minutes, shake the slide to remove the reagent.
9. Wash with distilled water for 5 minutes.
10. Dehydrate, clean, cover with xylene-based synthetic resin and examine under a microscope. |
