Ingredients |
A) Methanol fixative: Methanol, Formaldehyde, Fast Green FCF
B) Buffered xanthene solution: Eosin Y, Eosin B, Thiourea, Sodium Azide, Phosphate Buffers
C) Buffered thiazine solution: Methylene Blue, Azide A, Sodium Azide, Phosphate Buffers |
Working Principle |
It is particularly used to distinguish the nucleus and cytoplasm morphology of leukocytes, erythrocytes, platelets and parasites. One of its main uses is the staining of blood smears to look for Plasmodium spp in suspected malaria.
Giemsa stain is a classic blood film stain for peripheral blood smears and bone marrow samples. It stains erythrocytes pink, platelets pale pink, lymphocyte cytoplasm sky blue, monocyte cytoplasm pale blue and leukocyte nuclear chromatin magenta. |
Warnings, Precautions, Recommendations |
Hazard Highly flammable liquid and vapor. Toxic if swallowed, in contact with skin or inhaled. Causes skin irritation. Causes allergic skin reactions. Suspected of causing genetic damage. May cause cancer. Causes damage to organs. Keep away from heat/sparks/flame/hot surfaces. Do not smoke. Wear protective gloves/protective clothing/eye protection/face protection. Use personal protective equipment. IF INGESTED: Call NATIONAL POISON ADVICE CENTER TELEPHONE 114 or doctor/physician. IN CASE OF SKIN CONTACT: Wash with plenty of soap and water. IF INHALED: Remove the affected person to fresh air and keep in a comfortable position so that he/she can breathe easily. In case of exposure or interaction: Get medical advice/attention. Keep under lock and key. |

Blood Smears
1. Immerse and remove the air-dried smear 5 times for 1 second each in the “A-Methanol fixative”. Then shake the slide to remove the reagent.
2. Immerse and remove the preparation 5 times for 1 second each in the “B- Buffered xanthene solution” container. Then shake the slide to remove the reagent.
3. Immerse and remove the preparation 5 times for 1 second each in a beaker of “C- Buffered thiazine solution”. Then shake the slide to remove the reagent.
4. Wash with distilled water.
5. Air dry. (Do not use any heat source for drying). |
