Iodine, potassium iodide, 96% ethanol and PVP. |
Working Principle |
Designed for general laboratory use. Lugol's solution is a type of iodine solution. This solution is used to fix the crystal violet dye added in the previous step and the complexes formed in the bacterial cells. Lugol's solution forms the crystal violet-iodine complex, allowing it to bind more firmly to the bacterial cell wall. This staining procedure allows microorganisms to be divided into two groups. |

Application

The smear is usually fixed by heating.
If the smear is too acidic, cover with Sodium Hydrogen Carbonate Solution for 15-30 seconds.
Cover the smear with Crystal Violet Stain (GBL-2434) for 1-2 minutes.
Wash with distilled water.
Cover the smear with Gram Lugol's Solution (GBL-2435) for 1 minute.
Wash with distilled water.
Cover smear with Denatured Alcohol (GBL-2891) for 5-15 seconds.
Wash with distilled water.
Cover the smear with Safranin Stain (GBL-2444) or Gram Fuchsin Aqueous Stain (GBL-2424) for 1 minute.
Gently washed with distilled water. Air dried. Examined. |
