Purpose |
It is used to show Gram (+) and Gram (-) bacteria by light microscopy. |
Ingredients |
A- Gram Crystal Purple Solution: Crystal purple, ethanol, ammonium oxalate, dimethyl sulfoxide.
B- Gram Denatured Alcohol: Ethanol 96%, methanol, 2-propanol and stabilizer.
C- Gram Lugol Solution: Iodine, Potassium Iodide,96% Ethanol and PVP.
D- Gram Fuchsin Solution: Fuchsin, phenol and 96% ethanol. |
Working Principle |
Gram-Positive/Negative status is a characteristic that depends on the cell wall (peptidoglycan) structure of bacteria. The peptidoglycan structure of Gram-Negative bacteria is thinner shaped between the outer membrane and the inner membrane than Gram-Positive bacteria. In gram-positive bacteria, thick peptidoglycan has the property of retaining crystal purple. Lugol ensures that the paint is retained. Gram-positive bacteria have less cell wall permeability. Therefore, they do not release dyes during decolorization with denatured alcohol. In gram-negative bacteria, alcohol dissolves lipids in the outer membrane, the pores open and the crystalline purple dissipates. In this way, Gram-negative bacteria are stained with fuchsin, which is then added to the medium. |
Warnings, Precautions, Recommendations |
It is very flammable liquid and vapor. It leads to serious eye irritation. There is a suspicion of causing cancer. Keep away from heat/ sparks/flames/hot surfaces. Use protective gloves/ protective clothing/ eye protection/ face protection. In case of skin contact: Remove/Remove all contaminated clothing immediately. Dry your skin with water/ shower. In case of eye contact: Dry carefully with water for a few minutes. If it is attached and easy to do, remove the contact lenses. Keep rinsing. IF it is in case of exposure or interaction: Get medical help/care. Keep it under lock and key. |
Application
|
1. Cover the preparation with “A-Gram Crystal Purple Solution”. After 5 minutes, shake up and pour.
2. It is washed off carefully with distilled water.
3. Cover the preparation with “C-Gram Lugol Solution”. After 1 minute, shake up and pour.
4. It is washed off carefully with distilled water.
5. Cover the preparation with “B-Gram Denatured Alcohol Solution”. After 5-15 seconds, shake up and remove the reagent.
6. It is washed off carefully with distilled water.
7. Cover the preparation with “D-Gram Fuchsin Solution”. After 1 minute, remove the reagent by shaking off the microscope slide.
8. It is washed off carefully with distilled water.
9. Dry in the air, preferably on a heated flat surface. Examine it under a microscope.
