Application
|
Eosin Y, acetone and preservatives. Preparation of Randolph Dilution Solution:
Propylene Glycol 50 ml
Distilled Water 50 ml
1% Phloxine 10 ml
10% Sodium Carbonate 1 ml
Staining Method:
1. Capillary blood is drawn up to the 0.5 line of the leukocyte pipette.
2. Complete the volume with Randolph's dilution solution.
3. Shake vigorously for at least 2 minutes.
4. Fill the counting chamber (Thoma slide or Bright-Line slide) and wait 3-5 minutes.
5. Eosinophils appear red-pink.
6. All 16 large squares are counted in the Fruchs-Rosenthal counting chamber. |
