Working Principle |
Carbol fuchsin solution is used to bind to the cell walls of acid-fast bacteria. After staining, the preparations are treated with an acidic alcohol solution. The acidic alcohol removes the carbol fuchsin dye from cells other than acid-fast bacteria. This step ensures that only acid-fast bacteria remain stained, leaving other cells or structures clear. After decolorization, the preparations are treated with brilliant green dye. This dye stains the background and other cells other than acid-fast bacteria green. |
Warnings, Precautions, Recommendations |
Danger. Highly flammable liquid and vapor. Harmful if swallowed. Causes severe skin burns and eye damage. Suspected of causing genetic defects. Keep away from heat/sparks/flames/hot surfaces. Do not breathe dust/fume/gas/mist/vapors/spray. Wear protective gloves/protective clothing/eye protection/face protection. IF SWALLOWED: If you feel unwell, call the NATIONAL POISON CONSULTATION CENTER ON 114 or a doctor/physician. IF ON SKIN (or hair): Remove/take off immediately all contaminated clothing. Rinse skin with water/shower. IF IN EYES: Rinse cautiously with water for several minutes. Remove contact lenses, if present and easy to do. Continue rinsing. |
Application

1. The smear is stained with Carbol Fuchsin Stain by slowly heating the slide from the bottom using a burner or electric staining rack for 5 minutes until vaporization is observed. (Vaporization is continued for 3-5 minutes using low heat or intermittent heat.)
2. The slide is allowed to cool.
3. The slide is rinsed with water for 5-10 seconds.
4. The smear is covered with Acid Alcohol for 5-30 seconds.
5. It is washed very briefly with distilled water in a delicate manner.
6. The smear is stained with Brillant Green Stain for 1-2 minutes.
7. It is washed gently with distilled water.
8. It is dried in air.
9. The smear is examined with a 100 immersion objective. |
