Working Principle |
Gomori Methenamine Silver PASM Staining Kit is a staining kit used in histology to selectively highlight specific cell structures and tissue components, particularly basement membranes and fibrils. In this particular kit, the main components required to perform the Gomori silver Methenamine staining are Silver nitrate and Methenamine. Grocott Gomori's Methenamine silver is a special staining method more commonly used for fungi. The staining reaction is based on the fact that in the presence of periodic acid, the polysaccharides of the fungal cell wall are oxidized to aldehydes, which in turn reduce the nitrate-silver Methenamine complex due to the accumulation of reduced silver at the sites of the aldehydes, producing a brown to black color. |

Silver nitrate/Methenamine borate working solution:
Dissolve 1 cup of “C- Methenamine borate” mixture in 30 ml of “B-Silver nitrate solution” at room temperature. Allow it to dissolve completely. The mixture should be prepared freshly. Only glassware and plastic tweezers etc. should be used.
The mixture reaction starts at 55-57 0C. Bring the mixture to 55-57 0C using a hot water bath. The mixture should be used immediately and discarded.
1. Wash the preparation with distilled water for 2 minutes.
2. Soak in “A- Periodic acid solution” for 10 minutes.
3. Soak in distilled water 3 times for 30 seconds.
4. Soak in freshly prepared Silver nitrate/methenamine borate working solution for 35-45 minutes.
5. Soak in distilled water 3 times for 30 seconds.
6. Soak in “D- Gold chloride solution” for 1 minute.
7. Soak in distilled water for approximately 30 seconds.
8. Soak in “E- Sodium thiosulfate solution” for 2 minutes.
8. Wash under running water for 3 minutes.
9. Soak in distilled water for approximately 30 seconds.
10. Soak in F- “Light green SF solution” for 2-3 minutes.
11. Soak in distilled water for 30 seconds.
12. Soak in 70% ethanol for 1 minute.
13. Soak in 96% ethanol for 1 minute.
14. Soak in 100% ethanol for 1 minute.
15. Soak in 100% ethanol for 1 minute.
16. Wash with xylene. |
