Working Principle |
The basis of eosin nigrosin sperm staining is based on the principle that dead sperm take the dye in and appear stained because their membrane structure is disrupted. |
Warnings, Precautions, Recommendations |
Caution. Causes serious eye irritation. IN CASE OF CONTACT WITH EYES: Rinse carefully with water for several minutes. Remove contact lenses if fitted and easy to do so. Continue rinsing. If eye irritation persists: Seek medical advice/help. Wear protective gloves/protective clothing/eye protection/face protection. |
Application
|
1. Mix the semen sample thoroughly.
2. Mix 0.05 ml of semen sample and 0.05 ml of Eosin-Nigrosin stain in a test tube and wait 30 seconds.
(1 drop = 0.05 ml)
3. After 30 seconds, prepare a smear preparation on a slide by taking 1 drop of the suspension and allow to air dry.
4. Fix in water-free fixative medium.
5. Examine under a bright-field microscope at x1000 magnification and immersion oil.
6. Count stained (dead) or unstained (live) cells with a laboratory cell counter.
