Purpose It is used to demonstrate alpha-naphthyl acetate esterase activity in leukocytes by light microscopy.
Ingredients A) Citrate solution pH: 5,4: Sodium chloride, Chloroform, Triton X-100
B) FB blue dye: Fast blue RR salt

Working Principle Non-specific esterase leukocyte is an enzyme found in the cytoplasm of leukocytes (white blood cells). This enzyme hydrolyzes ester bonds, breaking down esters into their free acid and alcohol components. If leukocytes contain the enzyme esterase, the cells stain as black granulations.
Warnings, Precautions, Recommendations Harmful if swallowed, in contact with skin or inhaled. May be corrosive, irritating.
Application 1. Immerse the preparation in the “Fixative solution”. After 30 seconds, shake off the slide and remove the reagent.
2. Wash with distilled water for 1 minute.
3. Immerse the preparation in the “Incubation solution”. Incubate at 37 °C and protected from light. After 30 minutes, remove the slide and shake to remove the reagent.
4. Wash with distilled water for 2 minutes.
5. Stain the preparation with “G- Hematoxylin solution Gill-3” for 2 minutes. At the end of the time, shake the slide to remove the reagent.
6. Wash with distilled water for 15 seconds.
7. Air dry. Examine under a microscope. Use only aqueous covering medium if desired.
