Working Principle |
As reticulocytes continue to mature in the bone marrow, they still retain RNA within the cell. For this reason, reticulocyte staining usually contains a dye that stains RNA.
Brilliant Cresyl Blue is a type of dye that is often used for reticulocyte staining. This dye is specifically formulated to stain RNA within cells. When reticulocytes are stained with Brilliant Cresyl Blue, the RNA within the cell is clearly colored and the reticulocytes are easily recognizable.
In this way, reticulocyte staining makes it easier to visualize and count young red blood cells under a microscope. |

Application
|
1. Put 10 drops (0.5 ml) of “Reticulocyte Stain; Brilliant Cresyl Blue” (GBL-2405) in a small test tube.
2. Add 3 drops of blood. Mix. Let stand at room temperature for 15 minutes.
3. Spread on slide.
4. Dry in air.
5. Examine under microscope. |
